Description
Description
Tamoxifen – a non-steroidal anti-estrogenic agent with weak estrogenic properties. Its action is based on its ability to block estrogen receptors. Tamoxifen, as well as some of its metabolites with estradiol, compete for sites with cytoplasmic estrogen receptors in tissues of the breast, uterus, vagina, pituitary and tumors with levels elevated estrogen receptors. In contrast, the estrogen receptor-tamoxifen receptor complex will not stimulate DNA synthesis in the nucleus and will inhibit cell division, which leads to tumor cell regression and death.
Pharmacological effects
The mechanism of tamoxifen's anti-estrogenic effects (to prevent the formation or action of female sex hormones) is due to its ability to competitively bind to estrogen receptors (sections of the cell wall that interact with female hormones ) in the target organs and thus the formation of estrogenreteptornogo complex with endogenous ligands Preventing (interaction with receptors of biological substances produced in the body) – 17-beta-estradiol.
Indications
As an anti-tumor agent, tamoxifen is used in breast cancer in postmenopausal women (postmenopausal body condition is characterized by a decrease in the function of female gonads and, conversely, the development of genital organs), Cancer of the lining of the uterus (uterine mucosa), as well as in anovulatory infertility (infertility due to lack of maturation of eggs in the ovaries).
dosage
Affect yourself in a pill. For breast cancer received according to 20-40 mg 1-2 times a day, Endometrial cancer – 30-40 mg 1-2 times a day , with anovulatory infertility 10 mg 2 times a day for 4 consecutive days, from the 2nd day of menstruation. Subsequent 4-day cycles can be increased to a dose of 20-40mg twice daily.
Side effects
Possible side effects: gastrointestinal disturbances, dizziness, rash, hypercalcemia (increased calcium in the blood), thrombocytopenia (decrease in the level of platelets in the blood). It may be moderate metrorrhagia (uterine bleeding). In this case, reduce the dose. It was also noted the development of thrombosis (formation of a blood clot in a blood vessel). Retinal changes may occur at high doses.
