Pharmacological effects
The active ingredient Lasix – furosemide – has a diuretic effect, which is blocked by the reuptake of sodium and chloride ions in the ascending part of the loop of Henle. Lasix acts to a small extent on the tubules, unless this mechanism is associated with antialdosterone activity or inhibition of carbonic anhydrase. Furosemide (Lasix) promotes the excretion of potassium, magnesium, calcium.
When Lasix is given intravenously, the blood pressure may rise fast enough to reduce the pressure in the pulmonary artery and the bias pressure in the left ventricle. The diuretic effect is observed even after 5 min., a maximum reached in half the period of diuretic duration of about 2 hours.
If the diuretic effect of Lasix is administered, it begins within the first hour, reaches a peak after 1.5-2 hours, the duration of the running time about 7 hours.
Lasix is metabolized in the body and eliminated mainly in the urine in the form of a compound with glucuronic acid.
dosage
Dosage and dosage regimens are set individually depending on the degree of disruption of the value of the water-electrolyte balance of glomerular filtration. Then necessary, depending on the dose adjustment, the severity of the patient's disease, the size of urinary excretion. Usually the drug is administered in the form of a tablet, but if this is impossible or we are talking about urgent conditions, the drug is administered intravenously, the introduction of Lasix should not be less than 1.5-2 minutes.
With moderate severity of edema syndrome initially, the dose of Lasix is 20-80 mg or 20-40 mg orally intramuscularly or intravenously, with little effect the dose can be increased to 40 mg when administered orally and 20 mg when administered by Lasix injection. Dose increase cannot be earlier than 6-8 hours after receiving an initial oral dose no earlier than 2 hours after parenteral administration. A dose adjustment is made until the diuresis is sufficient. A single coordinated dose can therefore be administered once or twice a day. The maximum effect of Lasix can occur when the drug is administered 2-4 times per week.
For children, the dose depends on the child's weight and how Lasix is given. The initial dose of oral administration of 2 mg / kg, followed by administration by injection – 1 mg / kg. The dose can then be increased to 2 mg/kg orally and 1 mg/kg for parenteral administration. Dose increase cannot be earlier than 6-8 hours after receiving an initial oral dose no earlier than 2 hours after parenteral administration.
